抗體檢測通用ELISA操作流程（效價測定）
1.包被步驟：
1）用包被緩沖液配制1-10 μg/ml抗原溶液（現配現用）
2）向高結合力ELISA板（貨號80112）每孔加入100μl抗原溶液，覆以封板膜，室溫輕搖孵育2-6小時或4℃過夜
3）吸棄溶液，用洗滌緩沖液洗板2次
2.封閉步驟：
1）用封閉緩沖液母液配制工作液，每孔加入300 μl，室溫輕搖孵育2-6小時或4℃過夜
2）吸棄封閉液并在吸水紙上拍干，板子可立即使用或密封保存于4℃備用
抗體效價檢測：
1）用抗體稀釋液對免疫前/免疫后血清進行梯度稀釋（1:100至1:100,000），每孔加入100ul。覆以封板膜，室溫輕搖孵育30分鐘。
2）吸棄液體，用洗滌緩沖液洗板5次（每次200-300 μl，洗滌不充分會導致高背景）
3）用抗體稀釋液配制HRP標記抗兔二抗（1:5,000-1:20,000），每孔加入100μl（稀釋不當會導致顯色過弱或背景過高）
4）覆以封板膜，室溫輕搖孵育30分鐘
5）吸棄液體，洗板5次后拍干
6）每孔加入100 μl現成TMB底物溶液
7）覆以封板膜，室溫輕搖孵育至陽性孔出現藍色（最長15分鐘）
8）每孔加入100 μl終止液，藍色轉為黃色
9）450 nm波長下讀取吸光度
注：所有試劑需使用新槍頭及加液槽添加。除特異性抗原/抗體外，ELISA所需全部試劑均可通過試劑盒獲取。
注意事項：
1）本ELISA流程適用于針對純化抗原（蛋白質或多肽）產生的抗體的定性檢測，不適用于血清或其他體液中抗體濃度的定量測定。
2）本流程默認一抗來自兔源，若使用其他宿主動物需選用相應二抗。
